Purification of full-length huntingtin – trials and tribulations!

I am back from vacation and time spent on other projects so now I am back in the lab working on my HD project. Unfortunately, it has not been smooth sailing, I have had numerous issues making the full-length huntingtin sample in the lab.

The protein is consistently aggregating in the purification protocol but I managed to get it fixed (phew!) and think this is probably happening as the sample could be quite sensitive to environmental changes, in particular buffer pH shift.

None-the-less, very frustrating but I look forward to making the protein this week for future EM experiments with Susan Lea  in April, can not wait to get to Oxford and get in the lab! All of my trials and tribulations can be read on Zenodo and I will have more to share very soon!
