Previously I showed that, in my in vitro assay conditions, huntingtin binds a short piece of DNA. To see if this is relevant and specific, I have generated huntingtin-DNA complex samples which have been prepared for analysis by cryo-electron microscopy.
I have also tried to optimized the grafix procedure which cross-links and stabilizes the protein sample to increase the cross-linking of the protein molecules.
These experiments can be found on Zenodo here. I hope to look at these samples next week when I visit Susan Lea in Oxford, very exciting!
